Rockland offers Enzyme Substrates for reporter enzymes commonly used in biotechnology: horseradish peroxidase (HRP), calf intestinal alkaline phosphatase (CIAP or ALP) and ß-Galactosidase (BGAL). For each enzyme, substrates are available that generate either a soluble or insoluble colored product. Soluble substrates are useful to quantitate analytes in liquid phase assays like ELISA. Insoluble substrates are excellent to localize analytes in solid phase assays like western blotting (WB) and immunohistochemistry (IHC). Chemiluminescent substrates are available for horseradish peroxidase for ELISA and Western Blots.

Peroxidase enzyme substrates are used in colorimetric, fluorometric and/or luminescent assays. These reagents are acted on by the enzyme and undergo a measurable color change or emit light in the presence of an enzyme-antibody complex bound to specific analytes. Soluble peroxidase substrates for ELISA include TMBE and ABTS. Insoluble peroxidase substrates for immunoblotting (WB) or immunohistochemistry (IHC) include TMBM and DAB.

Chemiluminescent HRP substrates are extremely sensitive, nonradioactive, enhanced luminol-based chemiluminescent substrates for the detection of horseradish peroxidase (HRP). Chemiluminescent HRP substrates are designed for both western blotting (WB) and enzyme immunoassay (ELISA) use. Rockland offers two types of chemiluminescent HRP substrates that differ in sensitivity. FemtoMax™ Super Sensitive Chemiluminescent HRP Substrate detects femtogram (10-15) levels of antigen. PicoMax™ Sensitive Chemiluminescent HRP Substrate detects picoogram (10-12) l levels of antigen.

Alkaline phosphatase enzyme substrates offered by Rockland are used in colorimetric assays. ALP or CIAP substrates are specifically acted on by the enzyme alkaline phosphatase and undergo a measurable color change in the presence of an enzyme-antibody complex bound to specific analytes. pNPP is a soluble alkaline phosphatase substrate for ELISA. Insoluble alkaline phosphatase substrates for immunoblotting (WB) or immunohistochemistry (IHC) include BCIP/NBT and BCIP/TNBT.

Beta Galactosidase enzyme substrates offered by Rockland are used in colorimetric assays for immunology , microbiology and molecular biology. BGAL substrates are specifically acted on by the enzyme ß-galactosidase and undergo a measurable color change, or white to blue color change, in the presence of antibody-enzyme binding target proteins or cells. Often beta galactosidase is used as a reporter in screening of combinatorial libraries or to select colonies expressing recombinant proteins after cloning. OPNG is a soluble BGAL substrate for ELISA. XGAL is the classical insoluble beta galactosidase substrate for immunoblotting (WB) or immunohistochemistry (IHC). IPTG induces the expression of any lac+ containing vector, plasmid or gene fragment and can be used with X-GAL to detect active ß-galactosidase.
