NAG-1 Antibodies and Reagents

 

 

Non-steroidal anti-inflammatory drug (NSAIDs) activated gene (NAG-1) is a member of the transforming growth factor-beta (TGF-beta) superfamily. NAG-1 is also known as macrophage inhibitory cytokine-1 (MIC-1) or growth differential factor 15 (GDF15) or placental bone morphogenetic protein (PLAB), or prostate derived factor (PDF). NAG-1/MIC-1/GDF-15 plays an important role in inflammation, apoptosis and cancer development. Expression of NAG-1 protein increased in inflammation, injury and malignancy and by a number of anti-inflammatory and anti-cancer drugs.

The NAG-1 protein (Uniprot Q99988) is synthetized as a 308 amino acid polypeptide that encompasses a 29 amino acid signal peptide, a 167 amino acid pro-peptide and a 112 amino acid mature region (from amino acid 197 to amino acid 308). The mature protein is secreted as a disulfide-linked homodimer of 112 amino acids which is released from the pro-peptide after intracellular cleavage at an RXXR furin-like cleavage site. A GC polymorphism changes basic amino acid histidine (H) to acidic amino acid aspartic acid (D) at the position 6 of the mature NAG-1 protein. This polymorphism is widespread among the population. The decreased risk of both sporadic and familial prostate cancer is shown to be associated with the NAG-1 D variant.
  
To assist the research community for investigating the function of NAG-1/MIC-1/GDF-15 polymorphisms, especially as this change may correlate with prostate cancer, Rockland Immunochemicals has developed a suite of NAG-1 reagents - including highly specific polyclonal and monoclonal NAG-1 antibodies. NAG-1 antibodies conjugated to either biotin or horseradish peroxidase are also available to further facilitate NAG-1 protein detection and quantification.

Suggested Products for Common Assays

 

 

For sandwich ELISA (E) to capture NAG-1 protein use a pan-reactive capture antibody (p/n 600-401-B07) in combination with Biotin Conjugated or HRP Conjugated pan-reactive or isoform-specific anti-NAG-1 detection antibodies. Biotin conjugated detection antibody will yield excellent signal to noise ratio when used in combination with streptavidin peroxidase or avidin peroxidase.

 

 

For Western Blotting (WB) use Peroxidase Conjugated Anti-NAG-1 (Rabbit) Antibody for colorimetric substrate and chemiluminescent substrate detection. Use our blocking buffer to reduce background and to enhance sensitivity.

 

 

For Immunofluorescence Microscopy (IF) use Biotin Conjugated Anti-NAG (Rabbit, C-terminal) Antibody in combination with streptavidin fluorescein conjugate. This antibody will recognize all forms of NAG-1 variants.

 

 

For Immunohistochemistry (IHC) use Peroxidase Conjugated Anti-NAG-1 (Rabbit) Antibody. HRP conjugates can be used in combination with MaxTag™ DAB tablets for immunohistochemistry.

REAGENTS AND KITS FOR PROFILING NAG-1 POLYMORPHISM


Abstract

Expression of NAG-1 protein increased in inflammation, injury and malignancy and by a number of anti-inflammatory and anti-cancer drugs.  Two NAG1 polymorphisms with basic amino acid histidine (H) and aspartic acid (D) at the position 6 of the mature NAG-1 protein is widespread among the population. The decreased risk of both sporadic and familial prostate cancer is shown to be associated with the NAG-1 D variant. Rockland Immunochemicals have developed a suite of NAG-1 reagents  -  highly specific polyclonal and monoclonal NAG-1 antibodies and their antibodies conjugated to facilitate NAG-1 protein detection and quantification.
